The synthesis of oligonucleotides primarily employs the chemical phosphoramidite triester method and solid-phase support. Commonly used immobilized surfaces include Controlled Pore Glass (CPG) or Polystyrene (PS) beads. The loading capacity of Controlled Pore Glass (CPG) supports is limited, approximately 50-90 μmol/g. The PSphase-HDmicro solid-phase support launched by InSaS utilizes polymerization technology to synthesize fixed cross-linking PS microspheres, offering higher loading capacities (up to 250 μmol/g for RNA synthesis and 400 μmol/g for DNA synthesis). It also features superior swelling ratios, uniform coverage of surface groups, and good inter-batch stability.
